Yes there is one big difference. Eairly manifolds used a WCFB carb, I forgot the last year. The mounting suface of the manifold is squarded off for this carb. To use an AFB style carb you must use and adapter plate for mounting the carb to the manifold. The later manifolds were cast for the AFB and have "feet" added to the mounting surface. I've attached a photo of the manifold showing the feet for an AFB carb. The primary and secondary bores were also larger, P-1 7/16", S-1 19/32".
