Hi there,
I have to tow a 56 Chrysler New Yorker with the Powerflite trans. and wondered if I can do this with a tow dolly and the rear wheels on the ground and the drive shaft hooked up ?
The distance is 150 miles.

No....dropping the drive shaft is the best thing to do. Chrysler guidelines said it was OK for limited distances at lower speeds......but i wouldn't chance it for 150 miles. If that pic is your car you've got a real beauty!.......

My plan was to drive it here under it's own power but after the auction ended I talked to the owner. He suggested I didn't because the bias ply tires are over 10 years old and he hasn't driven the car more than about 50 miles in that amount of time.
I found a transporter that will do it for $200. Better safe than sorry ! I've had lightly used but old tires come apart in the past. Don't want to repeat that again. It's got the 354 Hemi !

As others here have said, towing wthout dropping the drive shaft is not recommended, especially for 150 miles. It's fairly simple to do that - just a few bolts really. I have towed my 1958 De Soto on a tow dolly from Pennsylvania to Tennessee in just that fashion and no harm was done. I would have preferred towing in an enclosed trailer, but that wasn't an option at the time. 

Beautiful car. The seller gave you some excellent advice about not trusting 10 year old bias ply tires.
