Some 20 years ago I experienced the same problem on the driver's door of our 300B.  I ended up removing the door panel (wasn't fun) within the car.  Finally had to drill through the inner door skin to access the latch.  Something inside had broken and that was the only way.  You are lucky if that is all it is as the latch assemblies for (4) door and plentiful compared to 2 door examples. 
 
I ended up taking (3) assemblies from an Imperial sedan donor car and making one.  It took all day to do the project and I don't want to go there again.
